I have beeen involved in the functional characterization of SLC4 transporters using measurements of intracellular pH  in the mammalian HEK expression system.  Proximal renal tubular acidosis is a disease that severely affects the ability of the proximal tubule to absorb bicarbonate. The electrogenic sodium bicarbonate cotransporter NBCe1-A plays an important role. We have characterized the importance of TM1 in NBCe1-A and have studied the functional abnormalities in transporters expressing natural NBCe1-A mutations.  These studies are aimed at defining the transport site.  We use various methodologies including cysteine scanning with MTS reagents to uncover the molecular transport characteristics of the cotransporter.
